Injectable Multi-Drug Loaded Hydrogels for Contraception

Without a liver first-pass reaction, injectable intravaginal hydrogels could deliver drugs systemically. Due to its wide surface area, rich blood supply, relatively high permeability of many medications, and self-insertion, the vaginal route has many benefits. This paper focuses on the contraceptive function of a four-arm star-shaped poly(D,L-lactic-co-glycolic acid)-b-methoxy poly(ethylene glycol) (4sPLGA-mPEG) injectable temperature-sensitive block of copolymer hydrogels as a carrier of three drugs. The cumulative release amounts of indomethacin (IMC), gestodene (GSD), and ethinyl estradiol (EE) from copolymer hydrogels could be regulated by adjusting the lactide/glycolide (LA/GA) mol ratio by investigating in vitro controlled release profiles through HPLC. In addition, the IMC, GSD, and EE in vitro release profiles were well in line with the Higuchi model. The acute toxicity of multi-drug copolymer hydrogels filled with different dosing contents was measured in vivo. The uterus was hydropic in the high-dose population on day 1 and ulcerated on day 5, followed by intestinal adhesion. No festering of the tissues was observed with respect to the middle dosage community and blood coagulum occurred on various days in the uterus. No major tissue necrosis was observed in the low dosage community. Finally, the antifertility tests confirmed the excellent contraceptive effect of hydrogels filled with the multi-drug. The above results showed that, as a multi-drug carrier, injectable copolymer hydrogel was promising as a novel form of contraception.
